On top of all these glaring flaws, the game is riddled with bugs and technical issues. It's evident that the writing team put minimal effort into creating a compelling and engaging story. The dialogue is clichéd and uninspired, with cringe-worthy one-liners and forced attempts at humor. The narrative is convoluted and poorly paced, leaving players feeling disconnected and uninterested in the characters and their motivations. It's clear that the developers prioritized quantity over quality when it came to designing the game world.įurthermore, the storyline and writing are an absolute trainwreck. The environments feel lifeless and generic, with no memorable landmarks or captivating aesthetics. The textures are low-resolution and lack detail, giving the game a dated and unappealing look. The graphics and visuals are equally underwhelming. The overall mechanics are convoluted, leaving players confused and frustrated rather than engaged and entertained. It's as if the developers didn't bother to playtest their own game before releasing it to the public. The controls feel awkward and imprecise, making even the simplest tasks a chore. ![]() ![]() ![]() It is clunky, unresponsive, and lacks any sense of polish or refinement. This game falls far short of its promises and fails to deliver on almost every level.įirst and foremost, let's talk about the gameplay. So we arrived on Grahams Road – alongside Peter Mellish, the executive director of the ATV Federation and volunteer Blair Campbell – with our managing partner, general managers from Centennial Auto-Sport & Tire in Summerside and Charlottetown, Centennial's director of communications, and our main media man. However, Ser637 phosphorylation reverses this process. 4 Accumulating evidence indicates that Ser616 phosphorylation promotes the translocation of Drp1 from the cytosol to the mitochondrial outer membrane. 3 Strikingly, phosphorylation of Drp1 plays a crucial role in Drp1 activity regulation. The contribution of active Drp1 is one of the important factors that affect the function of mitochondria. 2 Drp1 is required for mitochondrial fission and acts as a protein marker for mitochondrial dynamics. The proteins that participate in fission and fusion regulation are deemed “mitochondria-shaping” proteins, including dynamin-related protein 1 (Drp1), and mitofusins 1 and 2 (Mfn1 and Mfn2, respectively). 1 Mitochondrial dynamics are equilibrated by the opposing processes of fission and fusion in response to the microenvironment. Dysregulation of mitochondria is closely related to tumor initiation and progression. Mitochondria are multifunctional organelles and perform functions, including regulation of cell bioenergetics, calcium homeostasis, reduction-oxidation balance, and programmed cell death. Disruption of Drp1 could be a promising therapeutic strategy for LMP1-positive NPC. Our findings provide novel insight into the role of EBV-LMP1-driven mitochondrial fission in regulating Drp1 phosphorylation at serine 616 and serine 637. In addition, EBV-LMP1 regulates Drp1 through two oncogenic signaling axes, AMPK and cyclin B1/Cdk1, which promote cell survival and cisplatin resistance in NPC. Targeting Drp1 impairs mitochondrial function and induces cell death in LMP1-positive NPC cells. Furthermore, the protein level of p-Drp1 (Ser616) is related to the clinical stage (TNM stage) of NPC. A high level of p-Drp1 (Ser616) or a low level of p-Drp1 (Ser637) correlates with poor overall survival and disease-free survival. We show that EBV-LMP1 exhibits a new function in remodeling mitochondrial morphology by activating Drp1. The role of Drp1 in the oncogenesis and prognosis of EBV-LMP1-positive nasopharyngeal carcinoma (NPC) was determined in our study. However, the mechanism by which oncogenic stress promotes mitochondrial fission, potentially contributing to tumorigenesis, is not entirely understood. Mitochondrial dynamin-related protein 1 (Drp1)-mediated mitochondrial fission has an impact on the chemoresistance of cancers. Recently, mitochondrial fission has been demonstrated as a crucial mechanism in oncovirus-mediated carcinogenesis. Latent membrane protein 1 (LMP1) is a major Epstein–Barr virus (EBV)-encoded oncoprotein involved in latency infection that regulates mitochondrial functions to facilitate cell survival. 11/13/2023 0 Comments Navy and silver christmas tree![]() I like to stick to classic or simple colors that I can use year after year, so this typically means I stick to white or metallics, like silver and gold. I found these wood snowflakes at our local dollar store and decided to leave them as they were for a simple and natural look and just propped them up on the branches of the tree.Įach year, I usually add a few new baubles to my collection. To give the tree a bit of a whimsical feel, I always like to add a few large-scale decorations to our tree. Then I added in a few flocked branches to give it more of a snowy look. ![]() I started by adding gold mesh and burlap ribbon to give it some fullness and texture. And because of that, I wanted it to feel cohesive with these spaces so I decided to decorate it using metallics for a hint of sparkle and a few touches of navy and natural wood tones. Our tree sits proudly between our open concept dining room and living room, and it can be seen from every room on our main floor. As soon as I let go of those confines everything seemed to fall into place…starting with our Christmas tree. It was just decor, so I was determined to stick it out. I felt silly…I had put so much thought into decorating our home and purchased quite a few new things. But after an entire morning of getting nowhere, my husband Nick finally asked me what I was doing. So I tried to push past it and continue decorating our home as I had set out. I thought the pillows were jarring against our cream couch, and the garland felt childish. It was all coming together and I was loving each new item I purchased.īut when the day came to finally decorate our home, I felt unsure. I started collecting a few things here and there red and navy tartan pillows, red and white felt ball garland, cream napkins with a red ticking stripe. I wanted a traditional Christmas for our little one and I wanted it to feel fun and casual, nothing too fussy. So I thought about it and decided this was the year I would decorate our home in red and white. I would be scrolling through Pinterest or flipping through a magazine and stop in my tracks when I came across a home beautifully decorated for the Christmas season in crisp whites and pops of red.Įach one looked so festive, happy and bright. I was imagining a soft and inviting palette of whites and indigo and navy blues, with a few natural wood accents mixed in. We had recently completed some major changes in our living room and dining room. I was eager to decorate these new spaces for the holidays. Long before we had even gathered for Thanksgiving I was envisioning our holiday decor. I have a little story I want to share with you about how our holiday decor came about this year and the lesson I learned about always trusting your instincts. So I start thinking about colors and themes for our decor and planning holiday recipes we’d like to try. Once Thanksgiving has passed (as Canadians our Thanksgiving always falls much earlier than our American friends), I am ready to completely embrace the holiday season. 11/13/2023 0 Comments Ion controlling qlab oscYou may wish to remove the triggers for the cues inside the Group, or keep them in place. For example, to set selected cues to not loop and hold on their last frame, you would put the OSC cues for these two functions into a Timeline Group cue (start-all Group cue,) then assign a hotkey or MIDI trigger to that Group. You can combine these in Group cues and give the Group cue a hotkey to build up more complex actions. Here’s whats included in the example workspace: If you put all your editing hotkey cues in a separate cue list, you can simply disarm that list when you are not programming. Remember that batch commands are potentially dangerous, particularly when they can be triggered by simple key presses. ![]() Generally, it’s best to keep all your hotkey cues together in a separate cue list so that they don’t get confused for cues that are part of your performance. You can copy and paste any cues you might find useful into your own workspaces. The example workspace contains many useful OSC commands that will work in this way. The Network cue (OSC cue) will start, and the selected cue(s) will be set to loop. We can now select all the cues which we want to loop and hit ⇧L. The example workspace has a MIDI trigger listening for a note 60, note on with any velocity. If you have a remote control device that sends MIDI notes, you could also set a MIDI trigger for the cue. ![]() L is generally already in use, as a shortcut for the built-in command “load”, so we might use ⇧L (shift-L). To do this, we assign a hotkey trigger to the Network cue (OSC cue.) This is done in the Basics tab of the inspector in QLab 3, or the Triggers tab of the inspector in QLab 4 or QLab 5. So what we need to be able to do is trigger the cue while we have one or more other cues selected. Of course, if we run this cue as it’s shown in the screen shot, nothing will happen, because the only cue selected is the Network cue (OSC cue) itself. Here’s a screen shot of our OSC cue that will set the selected cues to loops: If you are typing an OSC string from memory then the general rule is, all letters are lower case except the first letter of any words that are joined to another without a space, like ‘infiniteLoop’ where only the ‘L’ is capitalized. IMPORTANT: OSC is case-sensitive, so you must enter OSC with the capital letters in the correct place. In the text field, type /cue/selected/infiniteLoop 1 to switch the loop on or /cue/selected/infiniteLoop 0 to switch the loop off.For QLab 3 or 4: in the Settings tab of the inspector, set the message type to “custom OSC message.” In QLab 5, the message type is defined by the patch.Create a Network cue (in QLab 3, it’s called an OSC cue.).To set up a hotkey which will set the selected cue to loop, we do the following: QLab 5 defaults to requiring a passcode for OSC messages, which you can configure here: In QLab 3, setting up that patch looks like this: By default, QLab workspaces have OSC patch 1 set as a “loopback”, i.e the OSC is sent to the Mac QLab is running on, which has a special address, localhost, and to the port number which QLab listens for OSC on, port 53000. But you can also send OSC messages from QLab to QLab itself. In QLab, OSC is often used to communicate with other pieces of equipment over a network, such as an ETC Ion or a Meyer Galaxy. You can create hotkeys to invoke any OSC command in the dictionary this example will focus on looping. The OSC dictionary for QLab found in the QLab manual: ![]() You can use similar OSC cues on hotkeys to control a huge range of parameters in QLab. QLab 4 and 5 allow some cue parameters to be edited on multiple cues simultaneously, and looping is indeed one of them, but the hotkey method is still useful because it can be invoked at any time, and you don’t have to be looking at the correct tab of the inspector to use them. Once you’ve created these cues, you can assign hotkeys to them which makes them behave almost like built-in functions just like copy ( ⌘C) and paste ( ⌘V). In QLab 3 and later, now you can also use OSC (Open Sound Control) which can be a bit easier to set up. ![]() In QLab 2, this could be done with a Script cue. But what if you frequently have large numbers of cues you want to set to loop? It is possible to create a cue that will can perform this action on all the cues that are selected. For instance, QLab 3 only allows you to turn looping on or off for one Audio cue at a time, using the checkbox in the Time and Loops tab of the inspector. One of the great things about QLab is that its capabilities can be extended beyond those included in the program. ![]() Select date_trunc('day', t.date) as date, max(t.temp) as temp …a possible solution could be to use a WITH clause (or Common Table Expression ) to extract the maximum temperature per day and then join back to the original data for getting all the timestamps, like so: test=> with ttt as ![]() If you have an equivalent data-set in a traditional SQL database like PostgreSQL… test=> select * from temperatures order by date $tempMax refers to the maximum temperature field determined in the previous grouping stageįrom the filtered docs in variable maxOccur, only the date field is mapped to the resulting array using $$maxOccur.date as the mapping expression.$$i.temp refers to the temperature field of all elements in the items array which has variable name i here. ![]() Note that $ is used to reference root document fields whereas $$ is used to reference variables.
